Circular Dichroism Spectropolarimeter

Using the Circular Dichroism Spectropolarimeter

CD Operating Instructions:

  • Make sure liquid nitrogen level is above E-Mark.
  • Turn "pressure build" valve on cylinder.
  • Turn "gas use" valve on cylinder. The pressure should always be 30 psi.
  • O2 indicator (tube with silica and sand) should be brownish black.
  • Open valves (blue) of snubber parallel to tube (these valves are below the comp.) first L.H.S. the R.H.S.
  • Wait for 20 min.
  • Turn lamp power on. See the N2 indicators (blue mark should coincide with the N2 indicator). When "Lamp ready" lights up, turn on the lamp.
  • Record lamp hour. Allow lamp to warm up for 30 min (for better baseline)
  • Turn CPU and Mac on. Type "yes" on PC to initiate the instrument.
  • Start "Star Stationary" on Mac
  • Use Quartz cuvettes and seal them with stoppers. Make sure there is no chemical residue in contact with the cuvette holder.

For Quitting:

  • Transfer your raw data to your disc. It is now safe to turn off the power switch for the CPU
  • Record the lamp hour. Turn off the power switch for the lamp
  • Wait for 10 min to allow nitrogen gas to cool down the lamp
  • Shut valves in reverse order
  • Before you leave, make sure that the work area is neat and tidy.
